Kapari Bitkisinden Moleküler Baskılama Yöntemi ile Yüksek Saflıkta Kuersetin Eldesi

Biyoaktif bitki özlerinden biri olan ve yurt dışından ithal edilen kuersetin, elma, çilek, çay, fındık, kapari gibi bitkilerde bulunan flavonoidlerden olup önemli gıda maddelerinden biridir. Kuersetin molekülünün antioksidan, antiinflamatuar ve antikanser gibi farmakolojik aktiviteler gösterdiği bilinmektedir. Antik zamanlardan bu yana kullanılmakta olan kapari bitkisi (Capparis spinosa) ılıman Akdeniz ikliminde yetişir ve ülkemizde geniş bir yayılış gösterir. Kaparinin kuersetin ve türevleri bakımdan zengin olduğu yapılan çalışmalarda ortaya konmuştur. Dolayısıyla, kapari bitkisi yüksek saflıkta kuersetin eldesine imkân tanımakta ve Türkiye?deki kapari üretimi göz önüne alındığında bu potansiyelin değerlendirilmesi gerekmektedir. Bu projenin amacı ilk kez moleküler baskılanmış katı faz ekstraksiyonu (MISPE) ile kapari bitkisinden yüksek saflıkta kuersetin molekülü üretilmesi ve elde edilen hammaddenin ülke içinde kullanılmasını sağlamak, bu şekilde ülkemizin dışa bağımlılığını azaltmaktır. Çalışma süresince öncelikle kuersetin ile fonksiyonel monomerler arasındaki etkileşim FTIR ve UV/Vis spektrofotometre ile incelenmiştir. Bu çalışmada sentez için en uygun monomerin 4VP ve uygun çözücünün DMSO olduğuna karar verilmiştir. Sentezlenen polimerler ile deney tasarımı çalışması yapılmış, en uygun monomer ve başlatıcı derişimleri sırasıyla % Cm = % 5,0, % Ci =% 3,0 olarak belirlenmiştir. Bu koşularla sentezlenen polimerlerin karaterizasyonları FTIR analizleri ile yapılmıştır. Farklı ekstraksiyon yöntemleri ile kapari bitkisinin ekstraksiyonları gerçekleştirilmiş, en çok kuersetin elde edilen yöntemin asit hidroizi içeren ekstraksiyon yöntemi olduğuna karar verilmiştir. Ekstrelerin içerisindeki kuersetin ve rutin varlığı ITK ve HPLC ile doğrulanmıştır. Çalışma sonucunda kuersetini tanıyan moleküler baskılanmış polimerler üretilmiş ve bu polimerlerin kapari ekstresindeki kuersetini diğer analoglarına oranla daha fazla tuttuğu sonucuna varılmıştır. Elde edilen bu polimerler kapariden kuersetin eldesinde kullanılacaktır.

Risperidone is a second generation anti-psychotic drug that is commonly preferred in the treatment of schizophrenia, attention deficit hyperactivity disorder and bipolar disorders. Risperidone-associated hepatic adverse effects have long been investigated and continues to be unclear. In this review, our aim is to establish the possible hepatic adverse effects linked to risperidone treatment in children and adolescent in terms of severities and reversibility. We performed literature search via electronic databases (PubMed, Embase and the Cochrane Library) in August 2019. Risperidone treatment may lead to alterations in liver function test (LFT), mostly asymptomatic and reversible with the discontinuation or normalizes with time. Furthermore, there are few cases reporting risperidone-induced cholestatic hepatitis and immunoallergic hepatitis. Although exact mechanism of action of risperidone on liver remains unknown, there are growing evidences that link alterations of fatty acid biosynthesis to hepatic adverse effects. We recommend physicians to take basal LFT prior to risperidone treatment and monitor LFT during treatment. Additionally, physicians should be aware that most alterations at LFT associated with risperidone use are asymptomatic and reversible. However, more comprehensive studies investigating risperidone-associated hepatic adverse effects should be performed to reach a definitive outcome. © 2020, Cyprus Mental Health Institute. Psychotraumatology and Dissociation: A Theoretical and Clinical Approach

The term “psychotraumatology” can be considered as a fundamental term which consists of the whole of research and studies related to the post-traumatic stress disorder. This discipline, of which rise can be thought as simultaneous with the emergence of the increase in the number of studies on the post-traumatic stress disorder, can under no circumstances be examined and explained without referring to the dissociative disorders which refer to the whole of the short and long-term effects of early chronic childhood traumas by its very nature. The main goal of this study is, thus, to consider the relationship between the field of psychotraumatology which involves the factors causing psychological trauma, the traumatic process itself and the outcomes of it with the post-traumatic stress disorder and dissociation, both of which are trauma-based clinical phenomena, in a historical dimension and various perspectives. In accordance with this goal, academic approaches in the recent psychotraumatological literature will be discussed in the context of trauma and dissociation.
